On the Determination of Creatinine and Creatine in Urine

چکیده

Ever since the introduction of the calorimetric method for the determination of creatinine, potassium bichromate, in half normal solution, has been used as the standard for measuring the color obtained in the reaction of creatinine with picric acid and sodium hydroxide. That the bichromate has been serviceable for the purpose is evidenced by the fact that no other standard has ever been proposed. Potassium bichromate is, however, by no means ideal as a general standard of measure for the color comparisons involved in creatinine determinations. For determinations in ordinary urines containing 7-15 mgm. of creatinine in a volume of 5-15 cc. it may be regarded as satisfactory. For more dilute urines (or creatinine solutions) obtainable in unlimited quantities a fair degree of accuracy can be obtained with the bichromate standard by using the proportions of picric acid and alkali recommended by Shaffer. The use of the rigid bichromate standard imposes distinct and wholly unnecessary limitations on the applications of a remarkably flexible analytical method, and now that pure creatinine compounds can be prepared from urine with very little work it is, I think, a mistake to continue the use of the bichromate except perhaps for purely routine determinations in the course of ordinary urine analysis. The technique described in this paper is based on the use of 1 mgm. of creatinine as a standard in the making of the color comparisons.

A stability-Indicating HPLC Method for Simultaneous Determination of Creatine Phosphate Sodium and its Related Substances in Pharmaceutical Formulation

The objective of the study was to develop a simple, specific and stability-indicating HPLC method for the simultaneous determination of creatine phosphate sodium (CPS) and its related substances in pharmaceutical formulation. Separation of creatine phosphate sodium from its major process impurities and degradation products was achieved on a Hypersil BDS C18 column (250 × 4.6 mm, 5 μm) with an a...

A microcolorimetric determination of creatine in urine by the Jaffe reaction.

In 1945, a method was described by Bonsnes and the writer (1) for the determination of urinary creatine which involved only the addition of picric acid to dilute urine solutions and heating in a boiling water bath. An 80 per cent conversion of creatine to creatinine was obtained. The method has now been modified to achieve 100 per cent conversion, on the basis of Clark and Thompson’s recent stu...

The Occurrence and Determination of Creatine in the Urine. By

Some months ago we began to find creatine with surprising frequency in the urine of different patients, some of whom had shown no creatine on previous occasions. The fact that in all cases in which creatine was found the determinations were made by Folin’s new method -conversion of the creatine to creatinine by boiling with picric acid-whereas the earlier determinations had been made by the old...

On the Preparation of Creatine, Creatinine and Standard Creatinine Solutions

In order to enlarge the scope of the calorimetric method for the determination of creatinine and creatine it is, I think, necessary to replace the bichromate standard with standard creatinine solutions, and in the next two papers will be described various adaptations of the method based on this principle. In connection with this work I have again gone over the subject of the preparation of crea...

Development of Ratiometric Fluorescent Biosensors for the Determination of Creatine and Creatinine in Urine

In this study, the oxazine 170 perchlorate (O17)-ethylcellulose (EC) membrane was successfully exploited for the fabrication of creatine- and creatinine-sensing membranes. The sensing membrane exhibited a double layer of O17-EC membrane and a layer of enzyme(s) entrapped in the EC and polyurethane hydrogel (PU) matrix. The sensing principle of the membranes was based on the hydrolytic catalysis...
